Protein induced lipid demixing in homogeneous membranes

چکیده
Specific lipid environments are necessary for the establishment of protein signaling platforms in membranes, yet their origin has been highly debated. We present a continuum, exactly solvable model induced local demixing membranes. The coupling between composition and thickness membrane induces domains around inclusions with hydrophobic mismatch, even temperatures above miscibility critical point membrane. qualitatively explains experimentally observed formation by anchoring reconstituted actin flat supported bilayers.Received 31 May 2021Accepted 14 September 2021DOI:https://doi.org/10.1103/PhysRevResearch.3.L042013Published American Physical Society under terms Creative Commons Attribution 4.0 International license. Electric field-induced critical demixing in lipid bilayer membranes.
Electric fields can induce lateral reorganization of lipids in fluid bilayer membranes. The resulting concentration profiles readily are observed in planar-supported bilayers by epifluorescence microscopy. When a fluorescently labeled lipid was used to probe the field-induced separation of cardiolipin from egg-phosphatidylcholine, an enhanced sensitivity to the electric field was observed that ...
متن کاملLipid demixing and protein-protein interactions in the adsorption of charged proteins on mixed membranes.
The adsorption free energy of charged proteins on mixed membranes, containing varying amounts of (oppositely) charged lipids, is calculated based on a mean-field free energy expression that accounts explicitly for the ability of the lipids to demix locally, and for lateral interactions between the adsorbed proteins. Minimization of this free energy functional yields the familiar nonlinear Poiss...
متن کاملBinding of peripheral proteins to mixed lipid membranes: effect of lipid demixing upon binding.
Binding isotherms have been determined for the association of horse heart cytochrome c with dioleoyl phosphatidylglycerol (DOPG)/dioleoyl phosphatidylcholine (DOPC) bilayer membranes over a range of lipid compositions and ionic strengths. In the absence of protein, the DOPG and DOPC lipids mix nearly ideally. The binding isotherms have been analyzed using double layer theory to account for the ...
متن کاملProtein attraction in membranes induced by lipid fluctuations.
The nonspecific lipid-mediated attraction between two proteins embedded in a bilayer membrane have been investigated for a model system using Monte Carlo simulations. We found two types of attraction with different regimes. A depletion-induced attraction in the range r < sigmaL, where sigmaL is the diameter of a lipid and r is the distance between the surfaces of the two proteins, and a fluctua...
متن کاملLipid-protein interactions in membranes.
The interactions of lipids with integral and peripheral proteins can be studied in reconstituted and natural membranes using spin label electron spin resonance (ESR) spectroscopy. The ESR spectra reveal a reduction in mobility of the spin-labelled lipid species, and in certain cases evidence is obtained for a partial penetration of the peripheral proteins into the membrane. The latter may be re...
